Just put the Fusion CD in. I get an error saying "Access to the path C:\Program Files (x86)\Powerteq\Update\AutoUploader.dll" is denied.... Can I just plug my Edge Evo into my truck? Or do I have to wait and get this problem resolved??

Yes, you can just plug the Edge into your truck. You only need Fusion to perform updates or to load custom tunes.
You might try right clicking the Fusion icon and then choose "Run as Administrator" to get by this problem though. I don't know if this will work, but it won't hurt to try.
