I believe that you should do it on the dyno, if it is a common turbo setup then you can get a preliminary tune so you can drive it to the dyno to get it done the right way.
Each truck has a different base tune, and you have to base all your changes off of that tune, so yes you can a more generic tune than a dyno tune to run, but if you want the safest and fastest tune you will need to get on the rollers.
plus with a dyno tune it will be automatically adjusted for your altitude and if you have a good person doing the tune, you can have it adjusted to your driving style.
